目的探讨小檗碱在高糖高胰岛素环境下对大鼠胃体环形肌收缩活动的影响。方法采用张力测定法,在正常、高糖或/和高胰岛素环境下观察小檗碱对三磷酸腺苷(ATP)、卡巴胆碱(CCh)诱导大鼠胃体环形肌收缩反应的影响。结果(1)小檗碱对ATP(3μmol.L-)1、CCh(0.3μmol.L-1)引起的大鼠胃体环形肌收缩有明显的剂量依赖性抑制作用。(2)高糖(15.6,31.2 mmol.L-1)或/和高胰岛素(50~200 mU.L-1)环境下,ATP、卡巴胆碱诱导大鼠胃体环形肌收缩活动与正常环境下相比无明显变化(P>0.05)。(3)高糖(31.2 mmol.L-1)或高胰岛素(200 mU.L-1)环境下,小檗碱对CCh引起收缩反应的抑制作用明显增强(P<0.05或P<0.01),对ATP引起收缩反应的抑制作用显著减弱(P<0.05)。结论小檗碱明显抑制ATP和CCh引起的大鼠胃体环形肌收缩反应。在高糖高胰岛素环境下,小檗碱对大鼠胃体环形肌收缩活动的影响有所改变,特别是增强了对CCh收缩反应的抑制作用,可能导致糖尿病胃肠功能障碍的进一步加剧。
Objective To investigate the effect of berberine on contractile activity of the gastric corpus ring muscle in rats with high glucose and high insulin concentration. Methods Tension assay was used to observe the effects of berberine on the contractile response of gastric myocytes in rats induced by adenosine triphosphate (ATP) and carbachol (CCh) under normal, high glucose and / or high insulin concentrations. Results (1) Berberine had a dose-dependent inhibitory effect on the contractile contraction of the gastric corpus striatum induced by ATP (3μmol.L-) 1 and CCh (0.3μmol.L-1). (2) Under the condition of high glucose (15.6, 31.2 mmol.L-1) and / or high insulin (50-200 mU.L-1), ATP and carbachol induced contractile activity of the gastric corpus ring muscle in normal rats There was no significant difference between the two groups (P> 0.05). (3) The inhibitory effect of berberine on the contractile response induced by CCh was significantly enhanced (P <0.05 or P <0.01) under high glucose (31.2 mmol.L-1) or high insulin (200 mU.L- Inhibition of ATP-induced contractile response was significantly attenuated (P <0.05). Conclusion Berberine obviously inhibits the gastric muscle contraction reaction induced by ATP and CCh in rats.
