目的:探讨菟丝子总黄酮(TFCC)对体外培养大鼠成骨细胞骨代谢的影响。方法:取新生SD大鼠的颅盖骨,采用改良的组织块法培养成骨细胞。应用MTT法、对硝基苯二钠基质动力学法(PNPP),放射免疫法及ELISA法分别测定TFCC0.01,0.1,1,10,100μg.L-1对成骨细胞增殖、碱性磷酸酶(ALP)活性、骨钙素(BGP)含量及Ⅰ型胶原(COL-I)分泌的影响。结果:TFCC在48 h和72 h能显著促进成骨细胞增殖并提高ALP活性;48 h时TFCC可促进细胞上清液中BGP和COL-I的分泌。结论:TFCC能够明显促进大鼠成骨细胞的增殖与分化,并通过提高COL-I的分泌影响骨代谢。
Objective: To investigate the effect of total cotyledons (TFCC) on bone metabolism in cultured rat osteoblasts. Methods: The calvaria of newborn SD rats were taken and osteoblasts were cultured by modified tissue block method. The effects of TFCC0.01, 0.1, 1, 10 and 100μg.L-1 on proliferation of osteoblasts, alkaline phosphatase (ALP) activity, osteocalcin (BGP) content and collagen Ⅰ (COL-I) secretion. RESULTS: TFCC promoted osteoblast proliferation and ALP activity at 48 h and 72 h, while TFCC promoted secretion of BGP and COL-I in supernatant at 48 h. Conclusion: TFCC can significantly promote the proliferation and differentiation of rat osteoblasts and affect the bone metabolism by increasing the secretion of COL-I.
