目的探讨“延肾胶囊”对残肾肾小球系膜细胞凋亡及其细胞外基质代谢的影响。方法Wistar大鼠120只,随机分为6组,A组为假手术对照组,B组为5/6肾切除;C,D和E组分别为肾切除+低、中、高剂量延肾胶囊;F组为5/6肾切除+肾衰宁。术后A,B两组给予等容积生理盐水灌胃。定量喂养,自由饮水。于术后15,90天测定各指标,每组每时相点观察10只大鼠。结果术后B组血浆血管紧张素Ⅱ浓度、肾小球系膜细胞增殖指数和凋亡指数、肾组织TGF-β1,CoⅣ,Fas,FasL积分光密度等指标显著高于A组。C,D和E组血浆血管紧张素Ⅱ浓度、肾小球系膜细胞增殖指数、肾组织TGF-β1和CoⅣ积分光密度高于A组,但显著低于B组,随着延肾胶囊剂量增加,上述指标有逐渐降低的趋势;肾小球系膜细胞凋亡指数、Fas和FasL积分光密度等指标显著高于A,B两组,随着延肾胶囊剂量增加,上述指标有逐渐升高趋势。F组血浆血管紧张素Ⅱ浓度、肾小球系膜细胞增殖指数、肾组织TGF-β1和CoⅣ积分光密度,低于B组,但高于D,E两组;肾组织Fas和FasL积分光密度等指标显著高于A和B组,稍低于D、E两组,肾小球系膜细胞凋亡指数与B组无显著差异,显著低于C,D和E3组。结论“延肾胶囊”能显著抑制5/6肾切除大鼠残肾血管紧张素Ⅱ和TGF-β1的高表达,增加Fas和FasL的表达,抑制肾小球系膜细胞过?
Objective To investigate the effect of “Yanshen Capsule” on the apoptosis of glomerular mesangial cells and the extracellular matrix metabolism of residual glomerular mesangial cells. Methods 120 Wistar rats were randomly divided into 6 groups. Group A was sham-operated control group, group B was 5/6 nephrectomy; group C, D and E were nephrectomized and low-, medium-, and high-dose Yanshen capsules, respectively. F group was 5/6 nephrectomy + renal failure. After surgery, A and B groups were given intragastric administration of normal saline. Quantitative feeding, free drinking water. Results The concentrations of plasma angiotensin II, glomerular mesangial cell proliferation index and apoptosis index, and TGF-β1, CoIV, Fas and FasL integrated optical density in group B were significantly higher than those in group A. Plasma concentrations of angiotensin II, glomerular mesangial cell proliferation, and TGF-β1 and CoIV integrated densities in group C, D, and E were higher than those in group A, but significantly lower than those in group B, with doses of Yanshen capsules. With the increase, the above indicators have gradually decreased; Mesangial cell apoptotic index, Fas and FasL integrated optical density and other indicators were significantly higher than the A, B groups, with the increase in the dose of Yanshen capsule, the above indicators have gradually increased. The apoptotic index of glomerular mesangial cells was not significantly different from that of group B, which was significantly lower than that of groups C, D, and E3. Conclusion “Yianshen Capsule” can significantly inhibit the expression of angiotensin II and TGF-β1 in the remnant kidney of 5/6 nephrectomized rats, increase the expression of Fas and FasL, and inhibit the glomerular mesangial cells from overgrowth.
