目的:探讨不同连作条件对于穿山龙的无机元素吸收及总皂苷含量的影响。方法:采用电感耦合等离子体质谱(ICP-MS)对不同连作条件下的穿山龙药材与根际土壤中17种无机元素的含量进行测定,射频功率1.3 k W,等离子体流量15 L·min~(-1),载气流量1.16 L·min~(-1),采样深度8.6 mm,雾室温度2℃,分析时间3 s,测量数3次,排风量5.0~7.5 m3·min~(-1)(4.7~8.1 m·s~(-1));循环水的要求为流速5 L·min~(-1),温度15~20℃,压力230~350 k Pa。利用紫外分光光度法测定穿山龙中总皂苷含量,检测波长457 nm,通过SPSS 17.0统计软件对实验数据进行相关性分析。结果:穿山龙在正茬2~4年时,药材与土壤的无机元素呈显著相关;从连作一茬1年开始无明显相关性,药材中总皂苷含量亦呈下降趋势。结论:穿山龙随着连作年限的增加,其总皂苷的含量随之下降,说明连作障碍对药材中总皂苷的含量积累造成了影响,但与无机元素的变化无明显相关,说明无机元素只是造成连作现象的因素之一。
Objective: To investigate the effects of different continuous cropping conditions on the absorption of inorganic elements and the content of total saponins in Chuan Shanlong. Methods: The contents of 17 inorganic elements in Rhizosphere Radix and rhizosphere soil were determined by Inductively Coupled Plasma Mass Spectrometry (ICP-MS) with RF power of 1.3 k W and plasma flow of 15 L · min ~ ( -1), the carrier gas flow rate was 1.16 L · min -1, the sampling depth was 8.6 mm, the temperature of the mist chamber was 2 ℃, the analysis time was 3 s, the number of measurements was 3 and the exhaust volume was 5.0-7.5 m 3 · min ~ 1) (4.7 ~ 8.1 m · s -1). The requirement of circulating water is 5 L · min -1, the temperature is 15 ~ 20 ℃ and the pressure is 230 ~ 350 kPa. The content of total saponins in Asteraceae was determined by ultraviolet spectrophotometry. The detection wavelength was 457 nm. The correlation analysis of the experimental data was carried out by SPSS 17.0 statistical software. Results: The results showed that there was a significant correlation between medicinal materials and inorganic elements in the soil during the 2 ~ 4 years of cropping. There was no significant correlation between the content of inorganic elements in the soil and soil, and the content of total saponins in medicinal materials also showed a decreasing trend. CONCLUSIONS: With the increase of continuous cropping, the content of total saponin decreased, indicating that continuous cropping obstacle affected the content accumulation of total saponins in medicinal materials, but had no significant correlation with the change of inorganic elements, indicating that inorganic elements only caused continuous cropping One of the factors of the phenomenon.
