本实验在石井畅方法的基础上进行改进,用改进的二羟基甲苯法测定RNA,二苯胺法测定DNA重复性好,回收率高(DNA达99.55±0.3%),当待测样品浓度RNA在10—60μg/ml、DNA在10一100μg/ml范围内可明显提高测量灵敏度。同时探讨了正常大鼠脾脏、肠系膜淋巴结、骨髓和外周血液的RNA与DNA含量参考值,为机体造血组织核酸代谢障碍时RNA与DNA的显色、定量分析提供了较好的测检方法。
This experiment was improved on the basis of the Ishii Chang method. RNA was determined using the modified dihydroxytoluene method. The diphenylamine method was used to determine the DNA reproducibility and the recovery rate was high (DNA was 99.55±0.3%). When the concentration of the sample to be measured RNA was 10-60μg/ml, DNA in the range of 10-100μg/ml can significantly improve the measurement sensitivity. At the same time, the reference values of RNA and DNA content in spleen, mesenteric lymph node, bone marrow and peripheral blood of normal rats were discussed, which provided a good method for the color and quantitative analysis of RNA and DNA in the hematopoietic tissue of the hematopoietic tissue.
