目的分析正常早孕妇女和稽留流产患者外周血及蜕膜组织中CD56+CD16-和CD56+CD16+自然杀伤(NK)细胞比率以及细胞表面整合素αvβ3的表达。方法分别采集15例正常早孕妇女和7例稽留流产患者外周血和蜕膜组织,Ficoll密度梯度离心法分离单个核细胞悬液,流式细胞术检测样本CD56+CD16-和CD56+CD16+NK细胞的比率以及其细胞表面αvβ3表达。结果 与早孕妇女外周血相比,蜕膜组织中CD56+CD16-NK细胞比率增高而CD56+CD16+NK细胞的比率却降低(P<0.01);正常早孕妇女及稽留流产患者外周血和蜕膜组织中CD56+CD16-和CD56+CD16+NK细胞的比率均无差异(P>0.05)。与正常早孕妇女相比,稽留流产患者蜕膜组织CD56+CD16-NK细胞表面αvβ3阳性表达率降低而其平均荧光强度(GM)值却增高(P<0.05)。结论 NK细胞及其所表达的整合素αvβ3在早孕期胚胎植入及胎盘形成过程中发挥重要作用,稽留流产患者NK细胞表面整合素αvβ3的表达异常,可能是稽留流产发生的免疫致病因素。
Objective To analyze the ratio of CD56 + CD16- and CD56 + CD16 + natural killer (NK) cells and the expression of integrin αvβ3 on peripheral blood and decidua in normal pregnant women and missed abortion patients. Methods Peripheral blood and decidua were collected from 15 normal pregnant women and 7 patients with missed abortion. The suspension of mononuclear cells was separated by Ficoll density gradient centrifugation. The samples of CD56 + CD16- and CD56 + CD16 + NK cells were detected by flow cytometry And its cell surface αvβ3 expression. Results Compared with peripheral blood of pregnant women, the rate of CD56 + CD16-NK cells in decidua increased and the percentage of CD56 + CD16 + NK cells decreased (P <0.01). In normal pregnant women and missed abortion patients, There was no difference in the ratio of CD56 + CD16- and CD56 + CD16 + NK cells (P> 0.05). Compared with normal pregnant women, the positive rate of αvβ3 on the surface of CD56 + CD16-NK cells in decidual tissue of missed abortion decreased and the average fluorescence intensity (GM) increased (P <0.05). Conclusion NK cells and their expressed integrin αvβ3 play an important role in embryo implantation and placenta formation in early pregnancy. Abnormal expression of integrin αvβ3 on NK cells in patients with abortion may be the immune pathogenic factors of missed abortion.
