目的观察延龄草总皂苷对结肠癌细胞SW-620迁移与侵袭,基质金属蛋白酶(matrix metallo-proteinase,MMP)2,9表达的影响。方法培养结肠癌细胞SW-620,加入不同浓度的延龄草总皂苷(5,10,15 mg/L),采用划痕实验,transwell侵袭实验观察延龄草总皂苷对SW-620迁移与侵袭的影响,以Western blotting方法检测细胞中MMP-2,MMP-9蛋白的表达。结果延龄草总皂苷可有效抑制SW-620的迁移与增殖。延龄草总皂苷可浓度依赖性地降低MMP-2,MMP-9的表达。结论延龄草总皂苷可有效抑制结肠癌细胞SW-620迁移与增殖,可能机制为其降低了MMP-2,MMP-9的表达。
Objective To investigate the effects of trillium saponin on the migration and invasion of colon cancer cell line SW-620 and the expression of matrix metallo-proteinase (MMP) 2,9. Methods Human colon cancer cell line SW-620 was cultured in vitro. Triterpenoids (5, 10 and 15 mg / L) were added into the colon cancer cells. Scratch and transwell invasion experiments were performed to evaluate the effects of trichosanthin on SW-620 migration and invasion Western blotting was used to detect the expression of MMP-2 and MMP-9 in the cells. Results Trillium total saponin can effectively inhibit the migration and proliferation of SW-620. Trillium total saponin can reduce the expression of MMP-2, MMP-9 in a concentration-dependent manner. Conclusion Trillium saponin can effectively inhibit the migration and proliferation of colon cancer cell line SW-620, which may be due to its reduced expression of MMP-2 and MMP-9.
