目的:建立水飞蓟宾葡甲胺在犬体内血药浓度的高效液相色谱检测方法。方法:在弱碱性条件下,血样用叔丁基甲醚进行液-液提取。结果:犬血浆中的成分对水飞蓟宾葡甲胺的检测没有干扰,且水飞蓟宾葡甲胺异构体得到很好分离。线性范围为0.1~5.0μg.mL-1,高中低3个浓度的平均绝对回收率为(94.2±6.12)%,方法回收率为(104.6±11.05)%。结论:该方法简单准确,灵敏度高,适用于水飞蓟宾葡甲胺体内药动学及临床药理学研究。
OBJECTIVE: To establish a method for the determination of silybin meglumine in dogs by high performance liquid chromatography. Methods: In weak alkaline conditions, blood samples were triturated with tert-butyl methyl ether. RESULTS: The composition of the canine plasma did not interfere with the silybin detection of silybin and the silybin isomer of silybin was well separated. The average absolute recovery was (94.2 ± 6.12)% in the range of 0.1 ~ 5.0 μg.mL-1. The average recoveries were (104.6 ± 11.05)%. Conclusion: The method is simple, accurate and sensitive. It is suitable for the pharmacokinetics and pharmacology of silybin in vivo.
