目的 观察电针关元、后三里穴对SAMP8小鼠外周血树突状细胞CD40、CD80分子表达的影响.方法 将30只SAMP8小鼠随机分为快速脑老化组(Control组)、电针组(EA组)、假电针组(ShameEA组),每组10只.取“关元”、双侧“后三里”穴电针处理,每周6次,共治疗4周.治疗结束,眼眶采血,经细胞培养后,光镜观察细胞形态,并用流式细胞术检测树突状细胞CD40、CD80分子表达的差异.结果 各组外周血单核细胞贴壁后,用含rhIL-4(1000 U/mL)、rhGM-CSF(1000 U/mL)、10%FBS的RPMI-1640培养液培养5d后,光镜下可见细胞表面有明显的树突状突起;经流式细胞术检测,与Control组相比,EA组树突状细胞CD40+CD80+共表达有显著差异(P<0.01).结论 电针关元、后三里穴与增强SAMP8小鼠免疫力及提高外周血树突状细胞CD40、CD80分子表达有关.“,”Objective To observe the effect of electro-acupuncture at Guanyuan and Housanli acupoints on the expression of CD40 and CD80 in dendritic cells (DC) from peripheral blood in SAMP8 mice.Methods 30 Senescence accelerated prone mouse P8 (SAMP8) mice were randomized into SAMP8 model group (control group),electro-acupuncture group (EA group),and shame electro-acupuncture group (Shame EA group),with 10 mice in each group.Electro-acupuncture at Guanyuan,Housanli points on both sides were taken,6 times per week,totally treated for 4 weeks.Blood samples were collected from orbit at the end of treatment.Cell morphology was observed under light microscope after cell culture.The difference of CD40 and CD80 expression in DC was detected by flow cytometry.Results After peripheral blood mononuclear cells of each group sticking wall,incubated in the RPMI-1640 culture media with rhIL-4 (1000 U/mL),rhGM-CSF(1000 U/mL) and 10% FBS for 5d,obvious dendritic protrusions was seen on the surfaces of dendritic cells (DC).By the flow cytometry detecting,compared with SAMP8 control group,the expression of CD40+CD80+ on dendritic cells (DC) in EA group was significantly different (P<0.01).Conclusion Electro-acupuncture at Guanyuan and Housanli acupoints in SAMP8 mice is related to strengthening the immune and improving the expression of CD40 and CD80 on dendritic cells (DC) from peripheral blood in SAMP8 mice.
