目的:研究大黄5种炮制品中2个蒽醌苷类成分含量的变化规律。方法:HPLC法同时测定芦荟大黄素-3-CH2-O-β-D-葡萄糖苷、大黄素-8-O-β-D-葡萄糖苷的含量,以Agilent TC-C18(2)为色谱柱,以四氢呋喃-1%冰醋酸溶液(25∶75)为流动相,检测波长410 nm,流速0.9 mL.min-1,柱温35℃。结果:芦荟大黄素-3-CH2-O-β-D-葡萄糖苷的测定范围为0.009 0~0.270 4μg,r=0.999 6,大黄素-8-O-β-D-葡萄糖苷的测定范围0.039 6~1.188μg,r=0.999 6;平均回收率分别为97.85%,97.19%。结论:大黄炮制过程中2个蒽醌苷类成分含量发生了明显变化,生片、醋片的含量没有显著性差异,酒片的含量较生片增加但不显著,熟片、炭片含量较生片下降明显。
Objective: To study the variation of two anthraquinone glycosides in five processed products of rhubarb. Methods: Simultaneous determination of emodin-3-CH2-O-β-D-glucoside and emodin-8-O-β-D-glucoside by HPLC was carried out on an Agilent TC-C18 column The mobile phase was tetrahydrofuran-1% glacial acetic acid (25:75). The detection wavelength was 410 nm and the flow rate was 0.9 mL · min-1. The column temperature was 35 ℃. Results: The determination range of aloe-emodin-3-CH2-O-β-D-glucoside was 0.009 0-0.270 4 μg, r = 0.999 6. The determination range of emodin-8-O-β-D-glucoside was 0.039 6 ~ 1.188μg, r = 0.999 6; the average recoveries were 97.85% and 97.19% respectively. CONCLUSION: The content of two anthraquinone glycosides in rhubarb processing changed obviously. There was no significant difference between the contents of green tea tablets and vinegar tablets. Green film decreased significantly.
