目的 研究 5 ,7双羟色胺 (5 ,7- DHT)对不同功能运动神经元 (Mn)群调质 CGRP配布式样的影响。方法 利用霍乱毒素 B亚单位结合胶体金逆行标记神经元复合免疫细胞化学的非荧光双标记方法。结果 大鼠比目鱼肌 (慢肌 ) Mn(SOL - Mn)群和趾长伸肌 (快肌 ) Mn(EDL - Mn)群 CGRP配布式样不同 ,即 SOL - Mn群含较高比率 CGRP样免疫反应 (CGRP- L I)阴性和弱阳性 Mn,EDL - Mn群含较高比率 CGRP- L I强阳性 Mn;于脑室注射 5 ,7-DHT 14d后 ,可明显引起 SOL - Mn群 CGRP表达增加 ,所有标记的 SOL - Mn都呈 CGRP- L I阳性 ,CGRP- L I强阳性的 Mn比率显著增高 (P<0 .0 0 1) ;而 5 ,7- DHT则未引起 EDL - Mn群 CGRP- L I反应强度的增高。结论 调质CGRP在 SOL - Mn群和 EDL - Mn群的配布式样差异除与其所支配肌肉的功能活动方式密切相关外 ,还受到脊髓 5 -HT传入信息的影响 ,SOL - Mn群可能比 EDL - Mn群接受更多的 5 - HT能信息。
Objective To investigate the effect of 5, 7-HT (5, 7-DHT) on the distribution of calmodulin (CGRP) in different functional motoneurons (Mn) Methods Cholera toxin B subunit combined with colloidal gold retrograde labeling neuronal complex immunocytochemistry non-fluorescent double labeling method. Results Compared with the CGRP distribution pattern of the soleus muscle (slow muscle) Mn (Mn - Mn) group and the extensor digitorum longum (EDL - Mn) group, the CGRP distribution pattern of the SOL - Mn group was higher (CGRP-LI) negative and weakly positive Mn and EDL - Mn groups contained high CGRP-LI positive Mn. After intracerebroventricular injection of 5, 7-DHT for 14 days, the expression of CGRP in SOL - Mn group was significantly increased. (P <0.01) of SOL - Mn positive for CGRP - LI and strong positive for CGRP - LI (P <0.01). However, the activity of CGRP - LI in EDL - Mn group was not induced by 5 and 7 DHT Increase. Conclusions In addition to the close correlation between the distribution pattern of CGRP in SOL - Mn group and EDL - Mn group and the functional activity pattern of muscle under their control, the CGRP of SOL - Mn group may be affected by 5 - HT information of spinal cord. - Mn group accepts more 5-HT energy information.
