目的确定丹参活血益脉丸中丹参酮ⅡA的提取方法,建立丹参活血益脉丸中丹参酮ⅡA的含量测定方法。方法 2014年12月采用不同提取溶剂、提取方法、提取时间确定丹参酮ⅡA的提取方法。采用高效液相色谱法测定丹参活血益脉丸中丹参酮ⅡA含量。色谱条件为色谱柱:Synergi Hydro-RP 80A(4.6×250 mm);流动相:甲醇-水(75:25),检测波长:270 nm,柱温:35℃。结果丹参活血益脉丸中丹参酮ⅡA的提取方法为甲醇回流提取1 h。丹参酮ⅡA在0.049 2~0.172 2μg范围内线性关系良好,平均回收率97.7%,RSD为1.41%。结论采用该方法提取丹参酮ⅡA结果准确,重现性好,专属性强,分离效果好,能有效控制丹参活血益脉丸的质量。
Objective To determine the extraction method of tanshinone ⅡA in DanShen Huoxue Yimai Pills, and to establish a method for the determination of tanshinone ⅡA in DanShen Huoxue Yimai Pills. Methods In December 2014, different extraction solvents and extraction methods were used to determine the extraction time of tanshinone ⅡA. Determination of tanshinone Ⅱ A in Dan Shen Huo Xue Yi Mai Pills by high performance liquid chromatography. The chromatographic conditions were as follows: Synergi Hydro-RP 80A (4.6 × 250 mm); mobile phase: methanol-water (75:25); detection wavelength: 270 nm; Results The extraction method of tanshinone ⅡA in Danshen Huoxue Yimai Pill was methanol reflux extraction for 1 h. Tanshinone Ⅱ A showed good linearity in the range of 0.049 2-0.1722 μg with an average recovery of 97.7% and a RSD of 1.41%. Conclusion The method of extracting tanshinone Ⅱ A has the advantages of accurate and reproducible results, strong specificity, good separation effect, and can effectively control the quality of Danshen Huoxue Yimai Pill.
