MTT法是近几年颇受欢迎的一种测定活细胞数的技术。其原理是:MTT分子中的四氮唑环可被活细胞线粒体酶分解成甲臜(formazan)结晶,后者经适当溶剂溶解后成蓝色,其显色程度可准确反映出活细胞数量。1983年,Mosmann首先报道这一方法。因其快速、简便、重复性好,可半自动操作,故使用逐年增多。国内外学者采用MTT法研究细胞增殖,试图在各自的领域中取代传统的复杂办法。本文就其在抗癌药物研究中的应用作一概述。
The MTT method is a popular technique for measuring the number of living cells in recent years. The principle is that the tetrazolium ring in the MTT molecule can be decomposed into formazan crystals by living cell mitochondrial enzymes, and the latter is blue after being dissolved in an appropriate solvent, and the degree of coloration can accurately reflect the number of living cells. In 1983, Mosmann first reported this method. Because of its rapidity, simplicity, repeatability, and semi-automatic operation, its use has increased year by year. Domestic and foreign scholars use MTT to study cell proliferation and try to replace traditional complex methods in their respective fields. This article gives an overview of its application in anti-cancer drug research.
