目的 观察丹参酮Ⅱ A对黑素瘤A375细胞CXCR7及VEGF表达的影响,探讨其抗肿瘤的作用机制.方法 体外培养黑素瘤A375细胞,经4组不同浓度丹参酮Ⅱ A(0,1,2,4mg/L)作用后,ELISA法检测培养液中VEGF浓度,Real time-PCR法及Western blotting法分别检测其对CXCR7和VEGF mRNA及蛋白表达的影响.结果 丹参酮Ⅱ A处理黑素瘤A375细胞48h后,培养液中VEGF浓度随丹参酮Ⅱ A浓度的升高而降低,CXCR7及VEGF mRNA及蛋白表达随丹参酮ⅡA浓度的升高而降低,差异具有统计学意义(P<0.05).结论 丹参酮Ⅱ A能抑制黑素瘤肿瘤细胞内VEGF及CXCR7的转录和蛋白的表达,提示丹参酮Ⅱ A可能具有潜在的抗肿瘤作用.“,”Objective To observe the influence of Tanshinone Ⅱ A on CXCR7 and VEGF expression in malignant melanoma A375 cells,and investigate its anti-tumor mechanism.Methods Cultured malignant melanoma A375 cells in vitro were divided into 4 groups to be treated with tanshinone Ⅱ A at different concentrations (0,1,2,4mg/L).VEGF concentration in the nutrient solution was detected by ELISA.The effects of CXCR7 and VEGF mRNA/protein expression were detected by Real time-PCR and Western Blotting respectively.Results A375 cells were treated with Tanshinone Ⅱ A for 48h.The concentration of VEGF in culture medium decreased with the increase of tanshinone Ⅱ A concentration,The expression of CXCR7 and VEGF mRNA/protein decreased with the increase of tanshinone Ⅱ A concentration,The difference was statistically significant (P < 0.05).Conclusion Tanshinone Ⅱ A can inhibit the transcription and protein expression of VEGF and CXCR7 in melanoma tumor cells,suggesting that tanshinone Ⅱ A may have potential anti-tumor effects.
