目的:研究甘肃产素花党参水煎剂抗皮肤衰老的作用和机制。方法:昆明种小鼠随机分为模型组、空白对照组、党参低剂量组和党参高剂量组、阳性对照组,经颈背部皮下注射5%D-半乳糖连续6 w,建立小鼠亚急性衰老模型,在光镜及电镜下分别观察党参水煎剂对D-半乳糖致衰老小鼠皮肤组织形态的影响;检测小鼠皮肤组织丙二醛(MDA)含量、脂褐素(LF)含量、超氧化物歧化酶(SOD)活性、ATP酶活性及羟脯氨酸(HYP)的含量。结果:党参低剂量组(5g/kg)及党参高剂量组(15g/kg)均可改善皮肤组织形态,光镜下,党参低剂量组、高剂量组皮肤真皮层显著增厚,纤维成分增多,排列较紧密,皮肤形态学结构较接近空白对照组;电镜下,党参低剂量组皮肤组织细胞可见轻度损伤,细胞器肿胀及少量空泡形成,党参高剂量组皮肤组织细胞结构完整,接近空白对照组皮肤组织细胞;MDA含量和LF含量降低,SOD活性及ATP活性升高,HYP含量升高。结论:甘肃产素花党参水煎剂能改善皮肤组织病理形态和自由基代谢失调的状况,对皮肤有一定的抗衰老作用。
Objective: To study the anti-skin aging effect and mechanism of Cilostazophylla Cilostazol in Gansu Province. Methods: Kunming mice were randomly divided into model group, blank control group, Codonopsis low dose group and Codonopsis high dose group, positive control group, subcutaneous injection of 5% D-galactose through the back of neck for 6 w, Aging model. The effects of Codonopsis pilosula on the skin tissue of aging mice were observed under light and electron microscopes respectively. The contents of malondialdehyde (MDA), lipofuscin (LF) , Superoxide dismutase (SOD) activity, ATPase activity and hydroxyproline (HYP) content. Results: The skin morphology was improved by low dose of Codonopsis (5g / kg) and high dose of Codonopsis (15g / kg). Under light microscope, the dermal layer of Codonopsis low dose group and high dose group were significantly thicker and the fiber content increased , The morphology of the skin was closer and closer to the blank control group. Under the electron microscope, the skin tissue cells of Codonopsis low-dose group showed mild injury, organelle swelling and a small amount of vacuoles. The skin tissue of Codonopsis high-dose group was intact and nearly blank Control group skin cells; MDA content and LF content decreased, SOD activity and ATP activity increased, HYP content increased. Conclusion: Codonopsis przewalskii Gansu decoction can improve the histopathology of skin tissue and the imbalance of free radical metabolism, and has some anti-aging effects on the skin.
