目的 探讨建立适用于临床工作的检测乳腺癌组织中血管内皮生长因子(VEGF)表达的方法.方法 免疫组化法.RT-PCR、Western blot、Dot blot分析VEGF在乳腺导管癌组织的表达.结果 VEGF阳性细胞主要为肿瘤细胞,间质也着色.VEGF蛋白质分子量为46KD,含三种异构体.Dot bolt见于53%癌组织,VEGF表达水平高于癌旁组织.结论 Dot blot可作为临床工作中检测VEGF的初筛方法.
Objective To establish a method for the detection of vascular endothelial growth factor (VEGF) expression in breast cancer tissues for clinical work. Methods Immunohistochemistry. RT-PCR, Western blot, dot blot analysis of VEGF expression in breast ductal carcinoma tissue. The VEGF-positive cells were mainly tumor cells with interstitial pigmentation. The molecular weight of VEGF protein was 46KD and contained three isomers. The Dot bolt was found in 53% of cancer tissues and the expression level of VEGF was higher than that in adjacent tissues. Conclusion Dot blot can be used as clinical work. Detection of VEGF in the primary screening method.
