目前分离血清高密度脂蛋白(HDL)最常用的方法是肝素-氯化锰沉淀法。此法虽简便,但当血浆甘油三酯浓度较高时会影响HDL分离。1979年Allen报道用聚乙二醇-6,000(PEG-6,000)沉淀分离正常人血清HDL。我们在此基础上进一步用此法分离人高脂血清和正常大鼠血清HDL获得满意结果。经琼脂糖电泳证明,最终浓度为10%的PEG-6,000适合人血清HDL分离,即使血浆甘油三酯和β脂
The most commonly used method of separating serum high density lipoprotein (HDL) is the heparin-manganese chloride precipitation method. Although this method is simple, but high plasma triglyceride concentrations will affect HDL separation. Allen reported in 1979 that normal human serum HDL was isolated by precipitation with polyethylene glycol-6000 (PEG-6,000). On this basis, we further obtained satisfactory results by using this method to separate human HDL from human hyperlipidemic serum and normal rat serum. As demonstrated by agarose gel electrophoresis, PEG-6,000 at a final concentration of 10% was suitable for human serum HDL isolation even though plasma triglycerides and beta-lipids
