本文利用氯胺T碘标技术对肺癌单抗(McAb 3D3)体外结合特性进行分析,McAb 3D3与肺腺癌细胞株L342的结合常数为3.2×10~8mol~(-1),每个L342细胞表面McAb 3D3的平均结合位点数为2.5×10~6个。RIA观察各种处理因素对L342细胞表面结合位点与~(131)I-McAb 3D3结合抑制率的影响,1%NaIO4、0.2%蛋白酶K、100%甲醇和100%丙酮的结合抑制率分别为72%、46%、14%和16%。结果提示,McAb 3D3所针对的3D3抗原是一种糖蛋白。
In this paper, we used chloramine T iodide standard technique to analyze the in vitro binding characteristics of McAb 3D3. The binding constant of McAb 3D3 to lung adenocarcinoma cell line L342 was 3.2 × 10 ~ 8mol ~ (-1). Each L342 cell The average number of McAb 3D3 binding sites was 2.5 × 10 ~ 6. RIA was used to observe the effects of various treatment factors on the binding inhibition of 131 I-McAb 3D3 on the surface of L342 cells. The inhibition rates of 1% NaIO4, 0.2% proteinase K, 100% methanol and 100% acetone were 72%, 46%, 14% and 16%. The results suggest that the 3D3 antigen against which McAb 3D3 is directed is a glycoprotein.
