以马桑内酯大鼠癫痫样行为发作、脑电图改变和离体海马脑片CA1区锥体细胞痫样放电为指标,观察L-型电压依赖性Ca2+通过阻断剂硝苯吡啶对其致痫作用的影响。结果发现:实验组与对照组相比较,大鼠癫痫发作推迟,程度减轻,脑电痫波出现潜伏期延长,波幅下降。离体海马脑片CA1区锥体细胞痫样放电被抑制,EPSP降低(P<0.05)。结果提示,马桑内酯可能通过激活L-型电压依赖性Ca2+通道,使神经细胞内游离Ca2+增多而致痫。
Epileptic seizures, electroencephalogram (EEG) changes and corneal epileptiform discharges (CA1) of hippocampal slices were used as indexes to investigate the effects of L-type voltage-dependent Ca2 + Effect of epilepsy. The results showed that compared with the control group, the seizures in the experimental group were delayed and the degree of euthanasia was relieved. The latency and amplitude of the EEG wave were prolonged. In isolated hippocampal slices CA1 pyramidal cells were suppressed, EPSP decreased (P <0.05). The results suggest that mayangloride may activate epilepsy by activating L-type voltage-dependent Ca2 + channels and increasing intracellular free Ca2 + in nerve cells.
