报告脑室注射(icv)生长抑素(SST)对大鼠痛阈和脑内单胺类递质的影响,采用Wistar大鼠作实验和病理观察。结果:Icv SST 5μg或IOμg可使大鼠痛阈升高,高效液相谱—电化学检测显示海马、下丘脑和脑干内5—HT和5—HIAA(5μg组脑干的除外)含量显著增多;而三脑区内NE含量的变化却不一致。icv SST 20μg时,高效液相色谱—紫外检测显示海马、下丘脑的次黄嘌呤和黄嘌呤明显下降,但是脑组织出现水肿。icv SST 40μg时,可见脑神经细胞核固缩等坏死性病理变化。提示有较严重毒性反应,临床用量不宜过大。
The effects of intracerebroventricular injection (icv) of somatostatin (Sv) on pain threshold and monoamine neurotransmitters in brain were reported. Wistar rats were used for experimental and pathological observation. Results: The pain threshold was increased in Icv SST 5μg or IOμg, and the contents of 5-HT and 5-HIAA in the hippocampus, hypothalamus and brainstem except for the 5μg group were significantly higher than those in the control group Increased; while the three brain area NE content changes are inconsistent. At 20 μg of icv SST, high performance liquid chromatography-ultraviolet detection showed a significant decrease in hypoxanthine and xanthine in the hypothalamus, but edema in the brain tissue. icv SST 40μg, we can see the necrotic pathological changes such as nuclear pyknosis of cranial nerve cells. Prompt more serious toxicity, clinical dosage should not be too large.
