目的采用RP-HPLC法测定大鼠血浆中的水飞蓟宾。方法以穿心莲内酯为内标,采用Kromasil C18色谱柱(150mm×4.6mm,5μm),流动相为乙腈-水(35∶65,10%磷酸调pH4.5),流速1.0mL·min-1,检测波长288nm,柱温35℃,进样量为20μL。结果水飞蓟宾0.5~100.0μg·mL-1与内标峰面积比的线性关系良好(r2=0.9999);高、中、低浓度的平均回收率分别为100.04%、99.82%、98.76%;日内和日间RSD均小于15%。结论所建方法简单、灵敏、准确可靠,可用于水飞蓟宾的药物动力学研究。
Objective To determine the content of silybin in rat plasma by RP-HPLC. Methods Using andrographolide as internal standard, the mobile phase consisted of acetonitrile-water (35:65, adjusted to pH4.5 with 10% phosphoric acid) at a flow rate of 1.0 mL · min-1 on a Kromasil C18 column (150 mm × 4.6 mm, , Detection wavelength 288nm, column temperature 35 ℃, injection volume of 20μL. Results The silybin 0.5 ~ 100.0 μg · mL-1 showed a good linear relationship with the internal standard peak area ratio (r2 = 0.9999). The average recoveries of high, medium and low concentrations of silybin were 100.04%, 99.82% and 98.76%, respectively. The RSDs are both less than 15% during and after the day. Conclusion The method is simple, sensitive, accurate and reliable and can be used to study the pharmacokinetics of silybin.
