目的:探讨制备小鼠膀胱整标本的方法.以及整标本技术在尿路上皮形态学中的应用。方法:取C57BL/6小鼠膀胱组织制作整标本.对制作完成的标本进行切片核染色和整标本核染色。用解剖显微镜.表面荧光显微镜和共聚焦荧光显微镜观察整标本膀胱的大体和超微形态.并建立小鼠膀胱上皮的3D成像。结果:小鼠膀胱整标本组织形态良好.膀胱上皮完整.表面平坦.黏膜皱襞全部消失。共聚焦荧光显微镜逐层扫描可观察到膀胱上皮全层及上皮下间质层。结论:小鼠膀胱整标本的制备简便易行.可弥补连续切片无法达到的组织整体观。可以用于组织形态学的分析研究.对于膀胱上皮发生学、生理学和病理生理学的研究具有十分重要的意义。
OBJECTIVE: To investigate the method of preparation of the bladder specimen in mice and the application of the whole specimen technique in urothelial morphology. Methods: C57BL / 6 mouse bladder tissue was taken to make the whole specimen, and the finished specimen was subjected to sectioned nuclear staining and whole stained nuclear staining. The gross and ultrastructural morphology of the whole bladder was observed with dissecting microscope, surface fluorescence microscope and confocal fluorescence microscope, and the 3D imaging of mouse bladder epithelium was established. Results: The integrity of bladder epithelium in mice was intact, the surface of bladder was smooth, the mucosal folds disappeared completely. Confocal fluorescence microscopy can be observed by layer-by-layer scanning of the full-thickness epithelium and the subepithelial stroma. Conclusion: The preparation of mouse bladder specimens is simple and easy to make, which can make up for the overall view of the tissue that can not be achieved by serial sections. It can be used for the analysis of histomorphology and is of great significance to the study of bladder epithelial genesis, physiology and pathophysiology.
