目的:探讨Gab2对结肠癌细胞MMP合成的影响。方法:临床选择2015年7月~2016年4月本院收治的接受手术的结肠癌病人的临床病理资料及组织标本,应用Western blot法检测接头蛋白Gab2水平,分析接头蛋白Gab2对结肠癌细胞侵袭相关蛋白MMP-2、MMP-9合成的影响。结果:结肠癌接头蛋白Gab2的阳性表达与病人的性别、年龄等临床指标无相关性,而与结肠癌的是否发生远处转移、肿瘤直径大小等临床指标密切相关,差异有统计学意义;接头蛋白Gab2阳性组的MMP-9、MMP-2指标水平显著高于Gab2阴性组,差异有统计学意义。结论:接头蛋白Gab2可通过影响结肠癌细胞MMP-9、MMP-2的表达进而影响结肠癌细胞的转移侵袭。
Objective: To investigate the effect of Gab2 on MMPs synthesis in colon cancer cells. Methods: Clinicopathological data and tissue samples from colon cancer patients undergoing surgery in our hospital from July 2015 to April 2016 were collected. The level of Gab2 was detected by Western blot and the effect of Gab2 on the invasion of colon cancer cells Related protein MMP-2, MMP-9 synthesis. Results: The positive expression of Gab2 in colonic carcinoma was not correlated with the clinical data such as gender and age of patients, but closely related to the occurrence of distant metastasis and size of tumor. The difference was statistically significant. The levels of MMP-9 and MMP-2 in Gab2 positive group were significantly higher than those in Gab2 negative group, the difference was statistically significant. Conclusion: The adapter protein Gab2 can affect the invasion and metastasis of colon cancer cells by affecting the expression of MMP-9 and MMP-2 in colon cancer cells.
