目的 应用原位末端标记技术对Balb/C乳鼠脑组织的凋亡细胞进行检测 ,并对脑组织中汉坦病毒 (HV)感染和细胞凋亡的关系进行初步探讨。方法 HV陈株经腹腔接种Balb/C乳鼠 ,感染后分别在 1、2、4、8、10和 14d取脑组织 ,4%多聚甲醛固定 ,常规制备 5 μm石蜡切片 ,应用原位杂交对乳鼠石蜡切片的病毒定位 ,用原位末端标记显示乳鼠脑组织中存在着凋亡细胞。结果 原位杂交检测结果显示 ,病毒主要累及大脑皮层、海马等部位 ,阳性信号主要分布于胞浆、紧靠核膜 ,原位末端标记的阳性细胞存在于神经细胞核内 ,在感染乳鼠脑组织中许多细胞被原位末端标记 ,呈胞核深染 ,弥漫分布于皮层、海马、白质等部位。结论 乳鼠脑组织中存在着凋亡细胞 ,该病变可能与病毒感染有关
Objective To detect the apoptotic cells in brain of Balb / C neonatal rats by in situ end-labeling and to investigate the relationship between Hantaan virus (HV) infection and apoptosis in brain tissue. Methods HV Chen strain was inoculated intraperitoneally with Balb / C suckling mice. After infection, the brain tissues were harvested at 1, 2, 4, 8, 10 and 14 days and fixed with 4% paraformaldehyde. Paraffin sections of 5 μm were prepared routinely. The virus localization of paraffin sections of suckling mice showed apoptotic cells in brain tissue of neonatal rats by in situ end-labeling. Results In situ hybridization results showed that the virus mainly involved in the cerebral cortex and hippocampus and other parts of the positive signals mainly in the cytoplasm, close to the nuclear membrane, the positive end-labeled cells present in the nucleus of the nucleus, infected neonatal brain tissue Many of the cells were labeled in situ, stained deep in the nucleus, diffusely distributed in the cortex, hippocampus, white matter and other parts. Conclusion Apoptotic cells exist in the brain of neonatal rats, which may be related to virus infection
