为了探讨低硒对鸡红细胞氧化应激和免疫功能的影响,在复制缺硒性渗出性素质鸡模型的基础上,采用实时荧光定量PCR等方法检测了红细胞脆性、抗氧化功能、细胞因子(IL-1β、IL-2、INF-γ和TNF-α)mRNA的表达量和红细胞免疫黏附促进因子(SOD1、SOD2、CD59和CR3)mRNA的表达量。结果表明:低硒组谷胱甘肽过氧化物酶(GSH-Px)活性显著降低,超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量显著升高,红细胞脆性增加,IL-2、INF-γmRNA表达量降低,IL-1β和TNF-αmRNA表达量升高,SOD1、SOD2的mRNA表达量降低,CD59、CR3的mRNA表达量呈上升趋势。说明缺硒可引起红细胞发生氧化应激,导致红细胞脆性增加,免疫功能障碍。
In order to investigate the effect of low selenium on the oxidative stress and immune function of chicken erythrocytes, real-time fluorescent quantitative PCR was used to detect the effects of erythrocyte fragility, anti-oxidative function, cytokines IL-1β, IL-2, INF-γ and TNF-α mRNA expression and the expression of erythrocyte immune adhesion promoting factor (SOD1, SOD2, CD59 and CR3) mRNA. The results showed that glutathione peroxidase (GSH-Px) activity was significantly decreased, superoxide dismutase (SOD) activity and malondialdehyde (MDA) content were significantly increased, erythrocyte fragility increased, IL- The mRNA expression of IL-1β and TNF-α increased, the mRNA expression of SOD1 and SOD2 decreased, and the mRNA expressions of CD59 and CR3 increased. Selenium deficiency can cause oxidative stress caused by red blood cells, resulting in increased fragility of red blood cells, immune dysfunction.
