背景:脑卒中易感型自发性高血压大鼠(SHRsp)是常用的高血压脑卒中动物模型。因其高血压使保种有一定难度,易受环境情况影响,从而使脑卒中特性易发生变异。目的:观察SHRsp的繁育能力和高血压脑卒中特性的保持。设计:以大鼠为观察对象,观察对比实验。单位:中国医学科学院阜外医院心血管病研究所。材料:实验于1999-09/2001-12在中国医学科学院阜外医院心血管病研究所进行。选择SHRsp种鼠93对186只,8周龄SHRsp大鼠48只和正常血压大鼠(WKY)98对196只进入实验。方法:采用兄妹交配方法,观察记录SHRsp2年的受孕率、孕鼠产仔数(仅计算未吃仔母鼠的产仔数),母鼠吃仔率,并与同期WKY大鼠比较;分别于12,16,20周龄时测量种鼠的收缩压和心率。另取48只8周龄SHRsp在10g/L盐水负荷下加速脑卒中的发生,自然死亡或盐负荷12周后断头处死,取大脑组织通过苏木精-伊红染色等处理后,在光镜下进行脑组织病理观察,检测脑卒中发生率。主要观察指标:2年雌鼠受孕率,孕鼠产仔率,母鼠吃仔率。种鼠收缩压,心率,脑卒中病理检出率。结果:SHRsp种鼠93对186只,WKY98对196只,48只8周龄SHRsp均进入结果分析。第1年度生产2代,平均SHRsp受孕率,孕鼠平均产仔数(100%,10.3只)高于同期WKY(90%,P=0.077;6.5只,P<0.001)。第2年度生产2代,平均SHRsp受孕率,孕鼠平均产仔数(89%,8.2只)高于同期WKY(59%,P<0.001;4.3只,P<0.001)。2年内SHRsp母鼠受孕87只,WKY母鼠受孕67只,SHRsp母鼠产仔后4周内吃仔率为6%(5/87),低于WKY吃仔率(18%,12/67),P>0.05。12周龄SHRsp雄鼠收缩压191.6~223.8mmHg(1mmHg=0.133kPa),雌鼠为174.2~196.3mmHg,16周龄雄鼠收缩压为219.0~224.9mmHg,20周龄雄鼠收缩压达232.0~242.6mmHg。SHRsp血压随周龄增加而升高。12周龄SHRsp雄鼠心率388~428次/min,雌鼠为373~417次/min。16周龄雄鼠心率392~410次/min,20周龄心率404~425次/min。48只雄鼠SHRsp脑卒中病理检出率为81%(39/48)。结论:SHRsp繁育能力与WKY相似。以血压、心率和脑组织病理检查保持了SHRsp高血压和脑卒中的特性,可做为高血压脑卒中的实验模型。
BACKGROUND: Stroke-prone spontaneously hypertensive rats (SHRsp) are commonly used animal models of hypertensive stroke. Due to its high blood pressure to maintain a certain degree of difficulty, susceptible to environmental conditions, so that stroke prone to mutation characteristics. Objective: To observe the breeding ability of SHRsp and the maintenance of stroke characteristics of hypertension. Design: The rats as the observation object, observe the comparative experiment. Unit: Fu Wai Hospital, Chinese Academy of Medical Sciences Institute of Cardiovascular Diseases. MATERIALS: The experiment was performed at Institute of Cardiovascular Diseases, Fu Wai Hospital, Chinese Academy of Medical Sciences from September 1999 to December 2001. A total of 186 pairs of SHRsp breeds, 488-week-old SHRsp rats and 98 of 196 normal-pressure rats (WKY) were enrolled in the experiment. Methods: By using sibling mating method, the SHRsp2-year-old male and female piglets were randomly divided into the control group and control group Sympathetic pressure and heart rate were measured at 12, 16 weeks of age. Another 48 8-week-old SHRsp accelerated the stroke under 10 g / L saline load. After natural death or salt stress for 12 weeks, the rats were decapitated and the brain tissue was treated with hematoxylin-eosin Microscopic pathological observation of brain tissue to detect the incidence of stroke. MAIN OUTCOME MEASURES: Pregnancy rate of 2-year-old female rats, litter size of pregnant rats, and eating rate of pregnant rats. Systolic blood pressure, heart rate, pathological detection rate of stroke. RESULTS: A total of 186 pairs of SHRsp breeds, 196 pairs of WKY98 and 48 8-week-old SHRsp were included in the analysis. The average annual SHRsp conception rate in the first year of production was higher than that of the pregnant WKY (90%, P = 0.077; 6.5, P <0.001). The average annual SHRsp conception rate in the second year of production was higher than that of the pregnant WKY (59%, P <0.001; 4.3, P <0.001). There were 87 pregnant rats and 67 pregnant WKY rats in SHRsp within 2 years, 6% (5/87) in 4 weeks after birth, which was lower than that of WKY (18%, 12/67 ), P> 0.05. SHRsp male rats at 12 weeks of age had systolic blood pressure of 191.6 to 223.8 mmHg (1 mmHg = 0.133 kPa), female rats of 174.2 to 196.3 mmHg, male rats at 16 weeks of age with systolic blood pressure of 219.0 to 224.9 mmHg, Systolic blood pressure of 232.0 ~ 242.6mmHg. SHRsp blood pressure increased with increasing age. The heart rate of 12-week-old SHRsp male mice ranged from 388 to 428 beats / min and that of female mice was 373 to 417 beats / min. 16-week-old male rat heart rate 392 to 410 beats / min, 20 weeks of age, 404 to 425 beats / min. 48 male rats SHRsp stroke pathological detection rate was 81% (39/48). Conclusion: The ability of SHRsp to breed is similar to WKY. Using blood pressure, heart rate and brain histopathology to maintain the characteristics of SHRsp hypertension and stroke can be used as an experimental model of hypertension stroke.
