目的:通过大鼠心肌缺血(寒瘀凝滞型)模型,观察三七吴茱萸流浸膏对大鼠心肌缺血(寒瘀凝滞型)的影响。方法:将60只WISTER大鼠分为6组,即:等体积生理盐水组,模型组,阳性组,大、中、小剂量组,每组10只,除生理盐水组外,其他各组大鼠每天饲以高脂饲料(含1%胆固醇,10%猪油,0.7%甲基硫氧嘧啶,88.8%基础饲料)[1],实验时间为28天,从第19天同时给药,阳性组每天1粒复方丹参片,大、中、小剂量组剂量分别为:10、5、2.5g/kg,每天1次。除生理盐水组以外,其余各组均从第25天开始在灌胃1h后,以2.5mL/kg的剂量腹腔注射异丙肾上腺素,并同时将大鼠放入0℃冰水中3min,每天1次,连续3天,最后一次注射24h后,用0.3%的戊巴比妥钠以30mg/kg剂量麻醉大鼠。麻醉后将大鼠仰窝放置,开胸取血(抗凝),用全自动血液流变仪测全血黏度(高、中、低切)、血浆黏度、红细胞压积、全血还原黏度(高、中、低切)、红细胞聚集指数、红细胞刚性指数、红细胞变形指数及红细胞电泳指数。结果:与模型组相比,各药物组均能显著改善血液流变学各项指标,且有显著性差异(P<0.01)。结论:吴茱萸三七流浸膏有良好的抗心肌缺血作用,能改善心肌缺血症状,有效的保护和治疗寒瘀型的心肌缺血。
OBJECTIVE: To observe the effects of Panax Notoginseng extract on myocardium ischemia (chilling stagnation type) in rats through a rat model of myocardial ischemia (Chilling stagnation). METHODS: Sixty WISTER rats were divided into 6 groups: equal volume saline group, model group, positive group, large, medium and small dose groups, 10 in each group. Except normal saline group, the other groups were large. Rats were fed daily with high-fat diet (containing 1% cholesterol, 10% lard, 0.7% methyl thiouracil, 88.8% basal diet) [1] for 28 days and administered simultaneously from the 19th day. One group of compound Danshen tablets per day, the doses of large, medium and small dose groups were: 10, 5, 2.5 g/kg, once daily. Except normal saline group, all other groups were injected intraperitoneally with isoproterenol at a dose of 2.5 mL/kg from the 25th day after intragastric administration. At the same time, the rats were placed in ice water at 0° C. for 3 min. At one time, rats were anesthetized with 0.3% sodium pentobarbital at a dose of 30 mg/kg 24 hours after the last injection for 3 consecutive days. After anesthesia, the rats were placed in an upside-down litter, and the thorax was opened for blood sampling (anticoagulation). The blood viscosity (high, medium, and low cut), plasma viscosity, hematocrit, and whole blood reduction viscosity were measured with an automatic hemorrometer. High, medium, and low cuts, erythrocyte aggregation index, erythrocyte rigidity index, erythrocyte deformability index, and red blood cell electrophoresis index. Results: Compared with the model group, each drug group could significantly improve the indexes of hemorheology with significant difference (P<0.01). Conclusion: Wuqi Sanqiliu extract has good anti-ischemic effect on myocardium, can improve the symptoms of myocardial ischemia, and effectively protect and treat cold-type myocardial ischemia.
