目的:建立HPLC法同时测定萝卜秦艽(Phlomis medicinalisDiels.)中sesamoside(Ⅰ)、山栀苷甲酯(Ⅱ)、dehydropentstemoside(Ⅲ)和8-乙酰基山栀苷甲酯(Ⅳ)等4种环烯醚萜苷含量的方法。方法:采用HPLC法,色谱柱:DiamonsilTMC18(250 mm×4.6 mm,5μm);流动相:乙腈-水,梯度洗脱;流速:1.0 ml/min;检测波长:235 nm;柱温:25℃。结果:4种环烯醚萜苷的线性范围分别为:Ⅰ:0.102~0.521 mg/ml(r=0.9999);Ⅱ:0.101~0.505 mg/ml(r=1.0000);Ⅲ:0.101~0.506 mg/ml(r=0.9999);Ⅳ:0.021~0.105 mg/ml(r=0.9998)。4种成分的平均回收率均在99.34%~100.23%,RSD≤1.5%。结论:本法简便、快速、准确,可以用来同时测定萝卜秦艽中4种环烯醚萜苷的含量。
Objective: To establish a method for the simultaneous determination of four cycloolefins such as sesamoside (I), methotrexate methyl ester (II), dehydropentstemoside (III) and 8-acetylcarboxidoside (IV) in Phlomis medicinalis Diels. The method of ether glycoside content. Method: HPLC method, column: DiamonsilTM C18 (250 mm×4.6 mm, 5 μm); mobile phase: acetonitrile-water, gradient elution; flow rate: 1.0 ml/min; detection wavelength: 235 nm; column temperature: 25° C. Results: The linear ranges of the four iridoid glycosides were as follows: I:0.102-0.521 mg/ml (r=0.9999); II:0.101-0.505 mg/ml (r=1.0000); III:0.101-0.506 mg/ Ml (r=0.9999); IV: 0.021 to 0.105 mg/ml (r=0.9998). The average recoveries of the four components ranged from 99.34% to 100.23% with RSD ≤ 1.5%. Conclusion: This method is simple, rapid and accurate. It can be used to simultaneously determine the content of four iridoid glycosides in G. radix.
