目的 :探讨采用流式细胞仪测定DNA含量及作细胞周期分析在诊断恶性体腔积液及判断腔内化疗疗效中的意义。方法 :对 41例患恶性肿瘤伴胸水、腹水和心包积液的患者 ,在腔内化疗前后进行积液中细胞DNA含量测定和细胞周期分析及细胞病理学检查。结果 :腔内化疗前 :41例患者中 19例查见癌细胞 ,3 4例中查见异倍体细胞。第一次腔内化疗 6~ 7天后 :41例患者积液中均未再查见癌细胞 ,有 3例查见异倍体细胞 ;DNA指数(DI)、G2 /M期所占比例 ,S期所占比例 (SPF)均显著下降 (P <0 0 0 1)。第二次腔内化疗 6~ 7天后 ,所有病例中均未查见癌细胞和异倍体细胞。结论 :采用流式细胞仪测定DNA含量及作细胞周期分析诊断恶性体腔积液和判断腔内化疗疗效具有快速、灵敏和可靠性高的优点
Objective : To explore the significance of determination of DNA content by flow cytometry and cell cycle analysis in diagnosing malignant effusion and evaluating the efficacy of intraluminal chemotherapy. Methods : Forty-one patients with malignant tumors with pleural effusion, ascites, and pericardial effusion were assessed for cell DNA content, cell cycle analysis and cytopathology before and after intracavitary chemotherapy. RESULTS: Prior to intraluminal chemotherapy: 19 of the 41 patients had cancer cells and 34 of them had aneuploid cells. After the first intraluminal chemotherapy 6 to 7 days: None of the 41 patients had oozes of cancer cells, and 3 had aneuploid cells; DNA index (DI), G2/M phase ratio, S The proportion of the period (SPF) decreased significantly (P < 0 0 01). The second intraluminal chemotherapy 6 to 7 days later, cancer cells and aneuploid cells were not found in all cases. Conclusion : The use of flow cytometry for the determination of DNA content and for cell cycle analysis has the advantages of rapidness, sensitivity and high reliability for diagnosing malignant effusion and judging the efficacy of intraluminal chemotherapy.
