目的:探讨瓜蒌薤白汤防治肺纤维化作用及机理。方法:采用一次性气管内注入平阳霉素3mg/kg诱导大鼠肺纤维化模型后,每天灌胃瓜蒌薤白汤,分别于第7天、14天后处死各组大鼠,观察大鼠肺泡炎、肺纤维化程度和肺泡灌洗液中超氧化物歧化酶(SOD)和肺组织中TNF-α、IL-10的表达。结果:第7、14天瓜蒌薤白汤组肺泡炎程度、肺组织TNF-α表达均显著低于模型组;第14天泼尼松对照组肺泡炎程度显著低于模型组,瓜蒌薤白汤组肺纤维化程度显著低于模型组。瓜蒌薤白汤组肺泡灌洗液中SOD活性、肺组织中IL-10的表达均高于模型对照组,但无统计学意义。结论:瓜蒌薤白汤能明显减轻平阳霉素所致的大鼠肺泡炎及纤维化程度,抑制肺组织中TNF-α过度表达。
Objective: To investigate the effect and mechanism of Gualoubai Decoction in prevention and treatment of pulmonary fibrosis. METHODS: Pulmonary fibrosis model induced by one-time intratracheal instillation of Pingyangmycin (3 mg/kg) was given to rats. The rats were sacrificed on the 7th and 14th day after the treatment. The rat alveolitis was observed. The degree of pulmonary fibrosis, superoxide dismutase (SOD) in lung lavage fluid, and the expression of TNF-α and IL-10 in lung tissue. Results: The degree of alveolitis and expression of TNF-α in lung tissue were significantly lower in the 7th and 14th days of the Gualoubai Decoction group than in the model group. The degree of alveolitis in the control group was significantly lower than that in the model group at the 14th day. The degree of pulmonary fibrosis was significantly lower in the group than in the model group. The activity of SOD in lung alveolar lavage fluid and the expression of IL-10 in lung tissue were higher in the Gualoubai Decoction group than in the model control group, but there was no statistical significance. Conclusion: Gualoubaitang can significantly reduce the degree of alveolitis and fibrosis induced by Pingyangmycin and inhibit the overexpression of TNF-α in lung tissue.
