辛伐他汀是羟甲基戊二酰辅酶A还原酶的特异性抑制剂,抑制内源性胆固醇的合成。近年的研究发现:辛伐他汀可促进成骨细胞增殖和骨形成增加,因此,学者们越来越关注该药在骨改建方面的应用。大量的研究表明:将不同载体携带的辛伐他汀局部应用于各种不同的动物模型可产生不同的促骨形成效果。辛伐他汀在体内应用促进骨形成的效果取决于局部的给药质量浓度,人们一直在努力寻找合适的给药途径。本文就辛伐他汀对骨再生所产生的影响、机制、载体和剂量等作一综述。
Simvastatin is a specific inhibitor of hydroxymethylglutaryl coenzyme A reductase, inhibiting the synthesis of endogenous cholesterol. Recent studies have found that simvastatin can promote osteoblast proliferation and bone formation, therefore, scholars pay more and more attention to the application of the drug in bone remodeling. A large number of studies have shown that the local application of different doses of simvastatin to different animal models can produce different bone formation-promoting effects. The effect of simvastatin on bone formation in vivo depends on the local drug concentration and people are always trying to find a suitable route of administration. This article reviews the effects of simvastatin on bone regeneration, its mechanism, carriers and dosages.
