目的 :探讨富血小板纤维蛋白(platelet-rich fibrin,PRF)对成骨细胞生物学特性的影响。方法 :体外培养成骨细胞MG63,实验组采用PRF,对照组为正常高糖DMEM培养液。以MTT法检测细胞增殖,碱性磷酸酶(ALP)染色法检测ALP活性,免疫组织化学法检测Ⅰ型胶原、骨保护素(OPG)及其配体(RANKL)的表达。采用SPSS17.0软件包对数据进行统计学分析。结果:PRF能够促进细胞增殖、ALP分泌、Ⅰ型胶原和骨保护素的表达,对骨保护素配体的表达影响不明显。结论:PRF能够促进成骨细胞增殖、分化及骨保护素的表达,PRF可能通过对骨保护素及其配体的调节,参与骨的重建过程。
Objective: To investigate the effect of platelet-rich fibrin (PRF) on the biological characteristics of osteoblasts. Methods: The osteoblasts MG63 were cultured in vitro. PRF was used in the experimental group and normal glucose DMEM culture medium was used as the control group. The cell proliferation was detected by MTT assay. ALP activity was detected by alkaline phosphatase (ALP) staining and the expression of type Ⅰ collagen, osteoprotegerin (OPG) and its ligand (RANKL) was detected by immunohistochemistry. SPSS17.0 software package for statistical analysis of the data. Results: PRF could promote cell proliferation, ALP secretion, type Ⅰ collagen and osteoprotegerin expression, and had no obvious effect on the expression of osteoprotegerin ligand. Conclusion: PRF can promote the proliferation and differentiation of osteoblasts and the expression of osteoprotegerin. PRF may participate in the process of bone remodeling through the regulation of osteoprotegerin and its ligand.
