目的检测SLARP蛋白免疫原性及能否成为疟疾疫苗潜在候选抗原。方法构建pc DNA3.1(+)/Py SLARP真核表达载体并表达,免疫小鼠并检测SLARP蛋白抗体及相关细胞因子水平。结果经pcDNA3.1(+)/PySLARP免疫后的小鼠血清中总IgG、IgG1、IgG2a抗体水平高于对照组,差异有统计学意义(P<0.01),并且小鼠脾细胞悬液细胞因子IFN-γ、IL-2、IL-4和IL-10水平也明显高于对照组,差异有统计学意义(P<0.01)。结论 PySLARP蛋白可以同时诱导体液和细胞免疫应答,具有一定的免疫原性,可以作为疟疾疫苗成分的一种候选抗原。
Objective To detect the immunogenicity of SLARP protein and its potential candidate antigen for malaria vaccine. Methods The pcDNA3.1 (+) / Py SLARP eukaryotic expression vector was constructed and expressed. The mice were immunized and the levels of anti-SLARP antibodies and related cytokines were detected. Results The levels of total IgG, IgG1 and IgG2a in the serum of mice immunized with pcDNA3.1 (+) / PySLARP were significantly higher than those in the control group (P <0.01), and the levels of cytokines The levels of IFN-γ, IL-2, IL-4 and IL-10 were also significantly higher than those of the control group (P <0.01). Conclusions PySLARP protein can induce both humoral and cellular immune responses at the same time and has certain immunogenicity as a candidate antigens for malaria vaccine components.
