目的:探讨14-3-3σ和P53蛋白表达在食管鳞癌和癌旁正常黏膜上皮、各级癌前病变中的变化特征及相关性.方法:采用免疫组织化学方法检测14-3-3σ和P53蛋白在60例不同分化程度食管鳞癌和癌旁正常食管黏膜上皮、各级癌前病变中的表达变化.结果:癌旁正常食管黏膜上皮中均可检测到不同程度的14-3-3σ蛋白表达(表达量8.22),随着癌前病变进展,14-3-3σ的表达率和表达量逐渐下降;低分化鳞癌中14-3-3σ的表达量最低(表达量0.45),完全失表达率为64%(7/11).P53蛋白在鳞癌中表达率最高67%(27/40),表达量为4.86,而只有25%(6/24)癌旁正常上皮表达P53蛋白(表达量0.42).14-3-3σ和P53蛋白的相关性分析表明二者呈显著负相关(P<0.05).结论:P53蛋白可能通过负调控14-3-3σ基因的表达参与食管癌变多阶段演进过程,14-3-3σ和P53可能成为食管癌早期诊断及高危人群筛查的分子指标,同时也可能成为食管癌生物预防和疗效评价的新靶点.
Objective: To investigate the changes and correlation of 14-3-3σ and P53 protein expression in esophageal squamous cell carcinoma and adjacent normal mucosal epithelium, precancerous lesions at all levels.Methods: Immunohistochemical method was used to detect 14-3-3σ and P53 protein in 60 cases of different esophageal squamous cell carcinoma and adjacent normal esophageal mucosa epithelial, precancerous lesions at all levels of expression changes.Results: adjacent normal esophageal epithelium can be detected in varying degrees of 14-3-3σ The expression of 14-3-3σ gradually decreased with the progression of precancerous lesions. The expression level of 14-3-3σ was the lowest (0.45) in poorly differentiated squamous cell carcinoma The positive expression rate of P53 protein in squamous cell carcinoma was 67% (27/40), the expression level was 4.86, but only 25% (6/24) adjacent normal epithelium expressed P53 protein (P <0.05) .Conclusion: P53 protein may be involved in esophageal carcinogenesis by negatively regulating the expression of 14-3-3σ gene The multi-stage evolution process, 14-3-3σ and P53 may be the molecular indicators of early diagnosis of esophageal cancer and screening of high-risk groups, but also may be esophageal cancer New targets and evaluate the efficacy of prevention.
