目的观察模拟低氧暴露条件对雄性SD大鼠睾丸机能的影响。方法随机将20只SD雄性大鼠分为两组,对照组置650 m海拔高度饲养,低氧组置模拟6000 m海拔高度低压氧舱饲养。饲养14天后脱颈处死大鼠,采集血液及组织标本,分别测定血清IL-1β和丙二醛(MDA)含量、睾丸匀浆组织过氧化氢酶(CAT)、谷胱甘肽过氧化物酶(GPx)活性。HE染色观察睾丸组织形态学变化。结果低氧组大鼠血清MDA、CAT及GPx的浓度或活性均较对照组显著升高(P<0.05);两组血清IL-1β浓度无显著差异(P>0.05)。与对照组比较,可见低氧组大鼠睾丸组织部分生精小管内初级、次级精母细胞变性、脱落、坏死,间质血管充血、扩张,间质细胞出现细胞肿胀及炎性细胞浸润。结论低氧低压暴露可能通过破坏睾丸组织内抗氧化系统稳态,导致大鼠睾丸生殖机能损伤。
Objective To observe the effects of simulated hypoxic exposure on testicular function in male SD rats. Methods Twenty SD male rats were randomly divided into two groups. The control group was housed at an altitude of 650 m. The hypoxia group was used to simulate the altitude of 6000 m. After 14 days of feeding, the rats were sacrificed and their blood and tissue samples were collected. The contents of IL-1β and malondialdehyde (MDA), the activities of catalase (CAT), glutathione peroxidase (GPx) activity. The histological changes of testis were observed by HE staining. Results The concentrations of MDA, CAT and GPx in the hypoxia group were significantly higher than those in the control group (P <0.05). There was no significant difference in serum IL-1β between the two groups (P> 0.05). Compared with the control group, the primary and secondary spermatocytes were degenerated, shed, necrotic, interstitial vascular congestion and expansion in interstitial tubules of testis tissue in hypoxic rats, and cell swelling and inflammatory cell infiltration in interstitial cells. Conclusion Hypoxia and hypobaric exposure may damage reproductive function of testis in rats by destroying the steady state of antioxidant system in testis.
