目的:为探讨LHSA作为肝细胞靶向剂交联“425”制剂抗日本血吸虫肉芽肿免疫效应的机理.方法;采用流式细胞仪分析T细胞亚群和脾脏中B淋巴细胞电泳对应用LHSA交联“425”制剂的日本血吸虫感染小鼠(BALB/c)进行细胞免疫和体液免疫功能测定,并与单一用“425”制剂和感染对照小鼠进行比较实验,LHSA交联“425”制剂应用后2wk,相当于感染后6wk,CD4/CD8淋巴细胞亚群的比值为2.45,显著高于后两组的1.92和1.75,同期脾脏中B淋巴细胞总频率达79%,高于单一用“425”制剂和感染对照动物的62.50%和64.50%,特异性抗体水平(OD均值)为0.56+0.05,也显著高于(均PM0.01)后两组的0.35±0.03和0.37±0.07应用后3~4wk(感染后7-8wk)时,CD/CD比值为1.21~2.06,显著高于后两组的1.32~1.59和1.48-1.70;B淋巴细胞数三组动物相近,但抗体水平0.66+0.06~0.74±0.10(OD均值)仍高出后两组的0.46±0.11~0.49±0.09和0.45±0.10~0.55±0.07.结论:LHSA交联“425”制剂应用于抗日本血吸虫卵肉芽肿病变,能使病变显著减弱,显著提高细胞免疫和体液免疫方面,阐明了LHSA交联“425”制剂诱导宿主产生明显的抗日本血吸虫卵肉芽肿病变“425”制剂免疫效应的机理,具有理论意义和?“,”To probe the immunoregulation mechanism of antigranuloma induced by LHSA conjugated “425”reagent, METHODS:The splenil T lymphtcyet phenotypes,B lymphocyte and specific antibody were detected during 2nd to 4th wk after BALB/c mice had been injected with targeting LHSA conjugated “425”reagent (Group Ⅰ), “425”reagent only (Group Ⅱ) and infected as control (Group Ⅲ). RESULTS: The ratios of CD/CD(2.45~ 2. 06 )were much higher than those in Group Ⅱ(1.92~1.59)and Group Ⅲ(1.75~1.70), the percentages of B lymphocyte in Group Ⅰmice (79~51. 30%)were much higher than those (62.50~50.16%)in Group Ⅱand (64.50~54. 10% ) in Group Ⅲ. The specific antibody levels (OD mean value) of sera from Groop I mice (0.56±0.05~0.74±0.10)were significantly higher than those from Group Ⅱ(0.35±0.03~0.49±0. 09) and Garoup Ⅲ(0. 37±0. 07~0. 55±0.07). The findings demonstrated that CD yalue, CD/CD ratio, percentage of B cell and antibody levels were associated with SEG size. CONCLUSION:This was the first time that the targeting LHSA conjugated “425”reagent were emploged to diminish SEG,and elucidated its immunoregulation mechnism of anti-SEG. This reseach improved basic thoery and will contribute to practice in the future.
