目的:探讨西妥昔单抗与树突状细胞(DCs)对头颈部鳞状细胞癌(HNSCC)的杀伤作用。方法:采用细胞因子rhIL-4、rhGM-CSF与rhTNF-α体外诱导扩增人外周血单个核来源的DCs,将DCs诱导的T淋巴细胞与西妥昔单抗联合作用于HNSCC,采用MTT法与流式细胞仪检测其对HNSCC杀伤率与凋亡率的影响,并用Western blot检测各组EGFR与pEGFR蛋白表达。结果:西妥昔单抗与DCs联合应用较其分别单独应用时,可明显增强对HNSCC的杀伤作用与凋亡作用,其差异有统计学意义(P<0.05),但是各组西妥昔单抗的蛋白表达无明显差异(P>0.05)。结论:西妥昔单抗与DCs两者具有协同作用,联合应用能明显增强其对HNSCC的杀伤作用。
Objective: To investigate the killing effect of cetuximab and dendritic cells (DCs) on head and neck squamous cell carcinoma (HNSCC). Methods: DCs derived from human peripheral blood mononuclear cells were induced by rhIL-4, rhGM-CSF and rhTNF-α in vitro. DCs-induced T lymphocytes and cetuximab were used in combination with HNSCC. MTT assay Flow cytometry was used to detect the effect of HNSCC on killing rate and apoptosis rate. Western blot was used to detect the expression of EGFR and pEGFR in each group. Results: The combination of cetuximab and DCs could increase the killing effect and apoptosis of HNSCC significantly compared with that of DCs alone (P <0.05), but the combination of cetuximab Anti-protein expression was no significant difference (P> 0.05). Conclusion: Both cetuximab and DCs have a synergistic effect, and the combined application can significantly enhance the killing effect on HNSCC.
