目的探讨胰岛素样生长因子-1(IGF-1)对人精子氧化损伤的保护作用。方法用H_2O_2处理人精子悬液建立氧化损伤模型,将精子悬液分为对照组、H_2O_2组、IGF-1+H_2O_2组。分别于培养后12 h、24 h,采用精子分析仪检测精子活力,Diff-Quik染色法进行精子形态学分析,精子染色质扩散试验检测精子DNA碎片率。结果在培养后12 h、24 h,与对照组相比,H_2O_2组精子活力及前向运动百分比均显著下降,DNA碎片率明显升高(P<0.05);IGF-1+H_2O_2组较H_2O_2组精子活力及前向运动百分比均显著升高,DNA碎片率明显下降(P<0.05);各组间正常形态精子比率无显著性差异(P>0.05)。结论 IGF-1对氧化损伤的精子具有一定的保护作用。
Objective To investigate the protective effect of insulin-like growth factor-1 (IGF-1) on human sperm oxidative damage. Methods The oxidative damage model was established by treating human sperm suspension with H 2 O 2, and the sperm suspension was divided into control group, H 2 O 2 group and IGF-1 + H 2 O 2 group. The sperm motility was detected by sperm analyzer at 12 h and 24 h after culture. The sperm morphology was analyzed by Diff-Quik staining, and the sperm DNA fragmentation rate was detected by sperm chromatin diffusion test. Results Compared with the control group, the sperm motility and the percentage of forward movement of H 2 O 2 group decreased significantly at 12 h and 24 h after culture, and the DNA fragmentation rate was significantly increased (P <0.05). Compared with H 2 O 2 group The percentage of sperm motility and forward movement increased significantly, DNA fragmentation rate decreased significantly (P <0.05). There was no significant difference in normal spermatozoa ratio between groups (P> 0.05). Conclusion IGF-1 has certain protective effect on oxidative damage of spermatozoa.
