目的:观察藻黄合剂(ZHM)对高糖环境下人肾小球系膜细胞(HMC)单核细胞趋化蛋白1(MCP-1)表达的影响,探讨其在糖尿病肾脏疾病(DKD)防治中的意义。方法:体外培养HMC并鉴定,血清药理学方法制备ZHM低、中、高剂量组,空白对照组及海昆肾喜阳性对照组大鼠含药血清,10%各组含药血清培养基分别孵育高糖作用下HMC 48h,另设低糖组作为空白对照。ELISA法和RT-PCR技术分别检测各组系膜细胞MCP-1蛋白及mRNA表达情况。结果:MCP-1蛋白及mRNA表达量在高糖组较低糖组明显升高(P<0.01);ZHM中、高剂量组MCP-1蛋白及mRNA表达量均低于高糖组,且具有剂量依赖性;ZHM中、高剂量组与海昆肾喜组比MCP-1蛋白及mRNA表达差异无统计学意义。结论:高糖可诱导HMC中MCP-1表达,而ZHM可通过抑制高糖条件下HMC中MCP-1蛋白及mRNA表达,发挥对DKD的防治作用。
OBJECTIVE: To observe the effect of ZHM on the expression of monocyte chemoattractant protein-1 (MCP-1) in human glomerular mesangial cells (HMC) under high glucose conditions and to investigate its effect on the prevention and treatment of diabetic nephropathy In the meaning. Methods: HMCs were cultured in vitro and identified. Serum pharmacology methods were used to prepare serum containing ZHM low, middle and high dose group, blank control group and Haqunzngxin positive control group rats, and 10% High glucose HMC 48h, another set of low-sugar group as a blank control. ELISA and RT-PCR were used to detect the expression of MCP-1 protein and mRNA in mesangial cells. Results: The protein and mRNA expression of MCP-1 in high glucose group was significantly higher than that in low glucose group (P <0.01). The protein and mRNA expression of MCP-1 in middle and high dose ZHM group were lower than those in high glucose group Dose-dependent. There was no significant difference in MCP-1 protein and mRNA expression between ZHM medium and high dose groups and Haikun Shenxian group. CONCLUSION: High glucose can induce the expression of MCP-1 in HMC. ZHM can inhibit the expression of MCP-1 protein and mRNA in HMC under high glucose condition.
