目的探讨槲皮素对大鼠肾上腺嗜铬细胞瘤PC12细胞的诱导分化作用。方法 MTT法检测神经生长因子(NGF)、不同浓度槲皮素(5、12.5、25、50μmol/L)对PC12细胞增殖活性的影响。分别以NGF、不同浓度的槲皮素(5、12.5、25μmol/L)诱导PC12细胞分化,以PBS作为对照组,诱导3 d后,显微镜下观察并记录PC12细胞分化情况,统计分析细胞分化率(轴索长度≥细胞直径的细胞百分数)、具有轴索的细胞数、细胞平均轴索数、梭形细胞比例等细胞分化的形态学参数。结果 MTT检测发现:50μmol/L槲皮素可明显抑制PC12细胞增殖(P<0.05)。与对照组比较,NGF组、槲皮素组(5、12.5、25μmol/L)的细胞均表现出显著分化特征(均P<0.05)。不同浓度槲皮素组(5、12.5、25μmol/L)诱导PC12细胞分化能力没有明显统计学差异。结论槲皮素可诱导PC12细胞分化,但没有明显剂量依赖性。
Objective To investigate the effect of quercetin on differentiation of adrenal pheochromocytoma PC12 cells. Methods MTT assay was used to detect the effects of nerve growth factor (NGF) and quercetin (5,12.5,25,50μmol / L) on the proliferation of PC12 cells. Differentiation of PC12 cells was induced by NGF and quercetin (5,12.5,25μmol / L) at different concentrations respectively. PBS was used as the control group. After 3 days of induction, the differentiation of PC12 cells was observed under microscope and the cell differentiation rate (Axonal length ≥ cell diameter of the cell percentage), with axonal cells, the average number of axonal cells, spindle cell ratio and other morphological parameters of cell differentiation. Results MTT assay showed that 50μmol / L quercetin significantly inhibited the proliferation of PC12 cells (P <0.05). Compared with the control group, the cells of NGF group and quercetin group (5, 12.5, 25μmol / L) showed significant differentiation (all P <0.05). There was no significant difference in the differentiation of PC12 cells induced by different concentrations of quercetin (5, 12.5, 25μmol / L). Conclusions Quercetin induces PC12 cell differentiation without significant dose-dependent.
