目的研究胰高血糖素样肽2(GLP-2)对肠缺血-再灌注(I-R)幼鼠肠屏障功能的保护作用。方法采用肠I-R模型,将24只雄性Wistar幼鼠随机均分为假手术(C组)、肠I-R损伤(B组)和肠I-R损伤+GLP-2保护(A组)三组。光镜下观察小肠黏膜形态学改变,检测肠系膜淋巴结的细菌菌落情况、肠黏膜丙二醛(MDA)、血清D-乳酸、内毒素和肿瘤坏死因子α(TNF-α)水平的变化。结果B组肠黏膜损伤最明显,Chiu′s评分、肠系膜淋巴结细菌菌落计数、肠黏膜MDA、血清D-乳酸、内毒素和TNF-α水平均显著高于C组(P<0.01);与B组相比,A组肠黏膜损伤明显减轻,且Chiu′s评分、肠系膜淋巴结细菌菌落计数、肠黏膜MDA、血清D-乳酸、内毒素和TNF-α水平显著降低(P<0.05)。结论GLP-2能够减轻幼鼠肠I-R损伤、保护肠黏膜屏障、减轻细菌/内毒素易位、减少氧自由基和炎性介质的产生和释放。
Objective To investigate the protective effect of GLP-2 on intestinal barrier function in intestinal ischemia-reperfusion (I-R) rats. Methods 24 male Wistar rats were randomly divided into three groups: sham operation (group C), intestinal I-R injury (group B) and intestinal I-R injury + GLP-2 protection (group A) using intestinal I- The morphological changes of intestinal mucosa were observed under light microscope. The changes of bacterial colony, intestinal mucosal MDA, serum D-lactate, endotoxin and tumor necrosis factor-α (TNF-α) were detected in mesenteric lymph nodes. Results The intestinal mucosa injury of group B was the most obvious. Chiu’s score, bacterial count of mesenteric lymph node, MDA, serum D-lactate, endotoxin and TNF-α in group B were significantly higher than those in group C (P <0.01) Compared with the control group, the intestinal mucosal lesion in group A was significantly reduced, and the Chiu’s score, bacterial count of mesenteric lymph nodes, MDA in intestinal mucosa, serum D-lactate, endotoxin and TNF-α were significantly decreased (P <0.05). Conclusion GLP-2 can reduce the intestinal I-R injury, protect the intestinal mucosal barrier, reduce bacterial / endotoxin translocation and reduce the production and release of oxygen free radicals and inflammatory mediators.
