目的:探讨一种新型免疫毒素LHRH-PE40与人结肠癌细胞系Lovo表面LHRH受体结合的特性,观察其对细胞增殖产生的抑制作用及检测细胞凋亡。方法:用125Ⅰ标记法检测LHRH-PE40与Lovo结合的特异性;MTT比色法检测LHRH-PE40对胃癌细胞的杀伤活性,流式细胞仪检测细胞凋亡情况。结果:人结肠癌细胞系Lovo细胞表面可见配基-受体特异性结合,LHRH-PE40对Lovo细胞的半数致死量为0.24 mg.L-1,0.1~10.0 mg.L-1LHRH-PE40作用于Lovo细胞,其凋亡明显(P<0.01)。结论:结肠癌细胞Lovo细胞表面存在LHRH受体,LHRH-PE40对结肠癌细胞的增殖状态有明显的抑制作用及促凋亡作用。
OBJECTIVE: To investigate the binding of LHRH-PE40, a novel immunotoxin, to LHRH receptor on Lovo human colon carcinoma cell line, and observe its inhibitory effect on cell proliferation and apoptosis. Methods: The specificity of LHRH-PE40 binding to Lovo was detected by 125 Ⅰ labeling method. The cytotoxicity of LHRH-PE40 to gastric cancer cells was detected by MTT colorimetric assay, and apoptosis was detected by flow cytometry. Results: Ligand-receptor specific binding was observed on the surface of human colon cancer cell line Lovo. The half lethal dose of LHRH-PE40 against Lovo cells was 0.24 mg.L-1, 0.1-10.0 mg.L-1LHRH-PE40 Lovo cells showed obvious apoptosis (P <0.01). Conclusion: The LHRH receptor is present on the surface of Lovo cells in colon cancer cells. LHRH-PE40 significantly inhibits the proliferation of colon cancer cells and induces apoptosis.
