目的探讨鹿茸多肽对辐射诱导神经母细胞瘤细胞调亡后B淋巴细胞瘤-2(B-cell lymphoma-2,Bcl-2)基因和Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)基因表达的影响。方法将神经母细胞瘤经X射线2 Gy照射5 min,诱导细胞凋亡后,用400和800 mg/ml鹿茸多肽溶液处理细胞,同时设未处理细胞作为对照组。采用RTPCR法检测Bcl-2、Bax基因mRNA转录水平。结果与对照组相比,鹿茸多肽400、800 mg/ml处理组Bcl-2、Bax基因mRNA转录水平均上调(P均<0.05);800 mg/ml处理组Bcl-2基因mRNA转录水平高于400 mg/ml处理组,而800 mg/ml处理组Bax基因mRNA转录水平低于400 mg/ml处理组,但两组间差异均无统计学意义(P>0.05)。结论鹿茸多肽对神经母细胞瘤细胞的凋亡具有明显的抑制作用,其抑制细胞凋亡的作用是通过上调Bcl-2表达、下调Bax表达实现的。
Objective To investigate the effect of antler polypeptides on the expression of Bcl-2 and Bcl-2 protein (Bcl-2-associated X protein, Bcl-2 and Bax) after radiation-induced neuroblastoma cell death ) Gene expression. Methods After neuroblastoma cells were irradiated with 2 Gy of X-rays for 5 min, the cells were treated with 400 and 800 mg / ml antler peptide solution. Meanwhile, untreated cells were used as the control group. RTPCR method was used to detect the mRNA transcription of Bcl-2 and Bax. Results Compared with the control group, the transcription levels of Bcl-2 and Bax mRNA in the antler polypeptide 400 and 800 mg / ml groups were up-regulated (all P <0.05). The transcription level of Bcl-2 mRNA in the 800 mg / 400 mg / ml, while 800 mg / ml Bax mRNA was lower than 400 mg / ml, but there was no significant difference between the two groups (P> 0.05). Conclusion The antler polypeptide has a significant inhibitory effect on the apoptosis of neuroblastoma cells, and its inhibitory effect on apoptosis is through the up-regulation of Bcl-2 expression and the down-regulation of Bax expression.
