应用细胞培养技术,NorthernBlot和图像细胞分析技术(ICM)研究钙对中国人乳癌细胞株BCaP-37的生长、增殖和基因表达的影响。生长曲线结果显示,钙(18.0mmol/L,36.0mmol/L)对细胞生长具有显著抑制作用(p<0.01)。基因表达结果显示,钙(12.0mmol/L)对癌相关基因EGFRmRNA的表达有抑制作用。测定ICM的四项指标包括DI值,S期百分比,总DNA含量及核面积。结果显示,两实验组(1.2mmol/L,12.0mmol/L)DI值分别为1.78和1.77,表现为恶性细胞的特征。其它三项指标均显著低于对照组(p<0.01,p<0.05)。综合评价ICM各项指标结果显示,钙对细胞的增殖具有显著抑制作用。
The effects of calcium on the growth, proliferation and gene expression of Chinese breast cancer cell line BCaP-37 were studied by using cell culture technique, Northern Blot and image cell analysis (ICM). Growth curve results showed that calcium (18.0 mmol/L, 36.0 mmol/L) had a significant inhibitory effect on cell growth (p<0.01). The results of gene expression showed that calcium (12.0 mmol/L) inhibited the expression of cancer associated gene EGFR mRNA. The four indicators for determining ICM included DI, S-phase percentage, total DNA content, and nuclear area. The results showed that the DI values of the two experimental groups (1.2 mmol/L, 12.0 mmol/L) were 1.78 and 1.77, respectively, which were characteristic of malignant cells. The other three indicators were significantly lower than the control group (p<0.01, p<0.05). Comprehensive evaluation of ICM indicators showed that calcium has a significant inhibitory effect on cell proliferation.
