目的 研究胸腺基质细胞的抗原提呈能力。方法 应用OVA特异的、受IAd分子识别限制的辅助T细胞杂交瘤(3DO.18.3)识别由IAd分子提呈的OVA的CNBr水解片段而被活化后产生IL2,测定IL2活性来分析胸腺基质细胞的抗原提呈作用。结果 IFNγ能促进MTEC1和MTSC4表达IAd分子,并促进MTSC4表达B71分子。经IFNγ作用后,MTEC1和MTSC4均有抗原提呈能力,MTSC4的抗原提呈能力较强。结论 抗原提呈能力的强弱,与TSC表达IAd和B71分子的水平相关,表明不同类型的TSC有不同的抗原提呈能力。
Objective To study the antigen presenting ability of thymus stromal cells. Methods The OVA-specific helper T cell hybridoma (3DO.18.3) restricted by recognition of IAd molecule was used to recognize the CNBr-hydrolyzed fragment of OVA presented by IAd molecule and then activated to produce IL2. IL2 activity was measured to analyze the effect of thymocyte Antigen presentation. Results IFNγ promoted the expression of IAd molecules by MTEC1 and MTSC4 and promoted the expression of B71 molecules by MTSC4. After IFNγ effect, MTEC1 and MTSC4 have antigen presenting ability, MTSC4 antigen presenting ability. Conclusions The strength of antigen presenting ability correlates with the expression of IAd and B71 in TSC, indicating that different types of TSCs have different antigen presenting ability.
