目的:研究健康人替硝唑血浆和唾液浓度的相关性,为临床用唾液浓度代替血液浓度提供依据.方法:8名男性健康志愿者分别po单剂量替硝唑片剂2.0g,采用高效液相色谱法(HPLC)测定替硝唑唾液和血浆浓度,并比较两种浓度各自计算的药物动力学参数.结果:替硝唑血浆浓度在1.13~44.9mg/L,唾液浓度(C_n)和血浆浓度(C_p)呈显著的正向相关关系(P<0.01).(C_n/C_p)为0.74±0.21(n=72).分别由C_n.和C_p,计算的药物动力学参数T_(1/2(K_a))、T_(1/2(k_e))、t_(peak)较为接近,无显著住差异(P>0.05),结论:可用唾液浓度代替血浆浓度进行替硝唑的药物浓度监测和人体药物动力学研究.
Objective: To study the relativity between the plasma and saliva concentration of tinidazole in healthy people and to provide the basis for replacing the blood concentration with clinical saliva concentration.Methods: Eight male healthy volunteers were treated with single dose tinidazole tablet 2.0g, The saliva and plasma concentrations of tinidazole were determined by HPLC and the calculated pharmacokinetic parameters were compared between the two concentrations.Results: The plasma concentrations of tinidazole ranged from 1.13 to 44.9 mg / L, saliva concentration (C_n) and plasma (C_p) was 0.74 ± 0.21 (n = 72), respectively.Compared with C_p and C_p, the calculated pharmacokinetic parameters T 1/2 (K_a), T_ (1/2) and t_ (peak) were close and there was no significant difference (P> 0.05) .Conclusion: The concentration of saliva can be used instead of plasma concentration to monitor the drug concentration of tinidazole and the body Pharmacokinetic studies.
