目的 :了解热休克转录因子 1(heatshocktranscriptionfactor 1,HSF1)基因对小鼠心肌组成型 (B晶体蛋白(αB Crystallin ,αBC)表达的影响。 方法 :用WesternBlot和免疫组织化学方法 ,测定组成型αBC在HSF1基因野生型(hsf1+/+)和HSF1基因敲除型 (hsf 1- /- )小鼠心肌中的表达。 结果 :αBC在hsf 1- /-和hsf 1+/+小鼠心肌表达量分别为 6 8.4 2 %± 4 .16 %和 10 0 %± 7.5 8% (心肌可溶性组分 ,P <0 .0 5 ) ,2 0 .5 3%± 1.0 1%和 37.5 5 %±1.91% (心肌不可溶性组分 ,P <0 .0 5 ) ;免疫组化显示αBC在hsf 1- /-心肌细胞内的表达信号较hsf1+/+明显减弱。结论 :hsf1基因是介导组成型αBC基因表达重要的、但不是唯一的因子。
OBJECTIVE: To investigate the effect of heat shock transcription factor 1 (HSF1) gene on the expression of α B crystallin (α B) in mouse myocardium.Methods: Western blot and immunohistochemistry (Hsf1 + / +) and HSF1 knockout (hsf 1- / -) mice.Results: The expression of αBC in the hsf 1- / - and hsf 1 + / + mice Were 6 8.4 2% ± 4.16% and 10 0% ± 7.5 8% (myocardial soluble fraction, P <0.05), 20.3% ± 1.0 1% and 37.5 5% ± 1.91% ( Myocardial insoluble fraction, P <0.05).
