目的 探讨针对端粒酶的反义寡核苷酸对喉癌细胞端粒酶活性的抑制及对P53和PCNA基因表达的影响。方法 用PCR ELISA法检测喉癌细胞株Hep 2细胞内端粒酶的活性。用免疫组化的方法检测喉癌细胞P53和PCNA基因的表达。结果 该反义寡核苷酸可抑制喉癌细胞端粒酶活性 ,此作用有序列特异性和浓度依赖性。 40 μM反义寡核苷酸抑制了喉癌细胞PC NA的表达 ,而对P53表达无明显影响。结论 靶向于端粒酶的反义寡核苷酸能抑制喉癌细胞Hep 2PCNA基因的表达。
Objective To investigate the effect of antisense oligonucleotide targeting telomerase on the telomerase activity and the expression of P53 and PCNA in laryngeal carcinoma cells. Methods The telomerase activity in laryngeal carcinoma cell line Hep 2 was detected by PCR ELISA. The expression of P53 and PCNA gene in laryngeal carcinoma cells was detected by immunohistochemistry. Results The antisense oligonucleotide can inhibit the telomerase activity of laryngeal carcinoma cells in a sequence-specific and concentration-dependent manner. 40 μM antisense oligonucleotide inhibited PCNA expression in laryngeal carcinoma cells, but had no significant effect on P53 expression. Conclusion Antisense oligonucleotides targeting telomerase can inhibit Hep 2PCNA gene expression in laryngeal cancer cells.
