目的:采用多指标综合评分法优选柴芍肠宁颗粒的提取工艺,为该制剂的生产工艺提供技术参数。方法:采用正交试验法,以出膏率和芍药苷、盐酸小檗碱、柠檬酸提取量为定量指标,小鼠小肠推进率、小鼠腹泻指数为药理指标,考察溶剂用量、提取时间、提取次数对提取效果的影响,通过综合评分法优选柴芍肠宁颗粒的提取工艺。结果:乌梅单煎的最优提取工艺条件为加10倍量水提取2次,每次1 h;柠檬酸提取量154.17 mg·g~(-1),出膏率31.27%。其余药味的最佳提取工艺条件为加10倍量水提取3次,每次1 h;芍药苷、盐酸小檗碱提取量分别为3.97,0.66 mg·g~(-1),出膏率26.36%,小肠推进率61.97%,腹泻指数0.98。结论:该提取工艺设计合理、稳定可行,对柴芍肠宁颗粒的生产工艺制定具有一定的指导意义。
OBJECTIVE: To optimize the extraction process of Chai Shao Chang Ning granules by multi-index comprehensive score method and to provide technical parameters for the preparation process. Methods: The orthogonal test was used to determine the amount of ointment, berberine hydrochloride and citric acid extract as quantitative indicators, mouse intestinal propulsion rate and mouse diarrhea index as pharmacological indicators. The effects of solvent dosage, extraction time, The effect of extraction times on the extraction efficiency was optimized. The extraction process of Chai Shao Chang Ning granules was optimized by the comprehensive score method. Results: The optimum extraction conditions for Fructus Pomona alone were as follows: extracting 10 times the amount of water and extracting twice each time for 1 h; extraction yield of citric acid was 154.17 mg · g -1, yielding an ointment of 31.27%. The best extraction conditions of the other flavors were extracted with 10 times the amount of water three times, each time 1 h; extraction of paeoniflorin and berberine were 3.97 and 0.66 mg · g -1, respectively, and the rate of ointment was 26.36 %, Intestinal propulsion rate of 61.97%, diarrhea index of 0.98. Conclusion: The extraction process is reasonable, stable and feasible, and has certain guiding significance for the formulation of Chai Shao-Ning Granules.
