很清楚,药物的剂型对药物在体内发挥疗效起到很重要的作用,特别是对于其血药浓度与效应有明确相关性的药物来说更是如此。二氢吡啶类钙通道阻断剂(CCB)就是这样一类药物。同为硝苯地平,但是如果剂型不同,它们的体内药动学过程以及降压疗效有极大差异,甚至可以说是完全不同。同样,不同长效CCB的体内药动学和交感神经激活方面的特点也有明显差异。对临床常用CCB的体内药动学过程进行分析,有助于我们识别真正长效的CCB类降压药物,为临床实践中合理的用药选择提供理论依据。
It is clear that the dosage form of the drug plays a very important role in the efficacy of the drug in the body, especially for drugs whose blood concentration and effect are clearly related. Dihydropyridine calcium channel blockers (CCB) is such a class of drugs. The same as nifedipine, but if the dosage forms are different, their pharmacokinetic process and antihypertensive efficacy are very different, or even completely different. Similarly, there are significant differences in the pharmacokinetics and sympathetic activation of different long-acting CCBs. The analysis of the pharmacokinetics of commonly used CCBs in clinical practice helps us to identify truly long-acting CCBs and provides a theoretical basis for rational drug selection in clinical practice.
